Tadalafil (C22H19N3O4), with a molecular weight of 389.4, has been widely used to treat erectile dysfunction (ED) in men since 2003, marketed as Cialis. In 2008, Eli Lilly sold its commercial rights for the treatment of pulmonary arterial hypertension (PAH) to United Therapeutics. In June 2009, the U.S. Food and Drug Administration (FDA) approved tadalafil for the treatment of PAH in the United States, marketed as Adcirca hypertension. Tadalafil was first marketed as an ED treatment in 2003. It takes effect 30 minutes after administration, with effects lasting up to 2 hours, and can last up to 36 hours, unaffected by food. The usual dose of tadalafil is 10 mg or 20 mg, with a recommended starting dose of 10 mg. Dosage adjustments should be made based on patient response and adverse reactions. Pre-marketing studies showed that after 12 weeks of oral administration of 10 mg or 20 mg of tadalafil, the response rates were 67% and 81%, respectively. Multiple studies have shown that tadalafil is effective in treating erectile dysfunction (ED).

【Specifications and Dosage】

Erectile dysfunction: This product is a film-coated tablet, available in 10 mg and 20 mg specifications. The recommended dose is 10 mg daily. If the 10 mg dose is ineffective, the dose can be increased to 20 mg. Take 0.5–12 hours before sexual activity.

Pulmonary Hypertension: 40 mg daily.
